DHEA Saliva ELISA kit relies on a competitive binding solid-phase enzyme-linked immunosorbent assay (ELISA). The kit uses antibodies coated on microtiter wells to bind to DHEA, both endogenous DHEA from a sample and DHEA conjugated to horseradish peroxidase (HRP). The amount of HRP conjugate bound to the antibodies is inversely proportional to the concentration of DHEA in the sample. After incubation, the unbound conjugate is washed away, and the bound conjugate is detected by adding a substrate that reacts with HRP, producing a color change. The intensity of the color is then measured, and the results are interpreted based on a standard curve.
