During the assay, the testos terone in the saliva samples competes with the enzyme-labeled testos terone for binding sites on the antibodies. The amount of enzyme-labeled testo sterone that binds is inversely proportional to the concentration of testos terone in the sample. After washing away unbound materials, an enzyme substrate is added, which reacts with the bound enzyme-labeled testos terone, producing a colored product. The absorbance of this product is measured, and the testo sterone concentration in the sample is determined by comparing the absorbance to a standard curve.
