The BI-EIA-GONADOTROPIN-LH is a one-step immunoassay, based on principle of sandwich method. The assay system utilizes a high affinity andspecificity monoclonal antibody (enzyme conjugated and immobilized) directedagainst a distinct antigenic determinant on the intact LH molecule. The testsample is allowed to react simultaneously with the two antibodies, resulting inthe LH molecules being sandwiched between the solid phase and enzyme-linkedantibodies. After incubation, the wells are washed with washing buffer toremove unbound labeled antibodies. A solution of TMB-Substrate is added andincubated, resulting in the development of a blue color. The color developmentis stopped with the addition of Stopping Reagent, changing the color to yellow.The concentration of LH is directly proportional to the color intensity of thetest sample. Absorbance is measured spectrophotometrically at 450 nm
